There is no strict adherence that certain people should not wear it. Garnet beads in the combination represent the 4 crimson-colored petals in this chakra. Regarding your second question, here is a short passage from the great book "Meditation & Mantras by Swami Vishnu-Devanananda" which I hope, will be of help: Muladhara Chakra "The Muladhara chakra is located at the base of the spinal column, It has a square mandala representing the earth principle which is a yellow in colour, and has the bija Mantra of Lam. The 4 petals which are crimson coloured are associated with the sound of vibrations of vam, sam, Sham and sum. These bijas begin on the upper right-hand petal and are read in a clockwise direction. Brahma is the presiding deity. Within this chakra, kundalini lies dormant. Here is also the Brahma granthi, or knot of Brahma, which must be forced open through rigourous sadhana and intense purification for the kundalini to rise. Meditation on the Muladhara chakra confers knowledge of the kundalini, as well as the means of awakening it. It bestows breath, and mind control, and knowledge of the past, present and future." According to the above information, the opening of Muladhara chakra does not usually led to rise of full kudalini but would rather be the begins of the next step of evolution, i.e. working on the next chakra up. In his book, the Sri Swamiji also mentioned guidance by a competent and experienced guru should be preferable. I tried to find the link for the book at Rudra Centre's website but could not locate it.
